Animal Imaging and Analysis (La Jolla)

Overview

The animal imaging and analysis lab provides state-of-the-art imaging and analytical services for Sanford-Burnham investigators. The facility can perform in vivo non-invasive luminescence and fluorescence imaging for xenograft tumor growth and metastasis studies. Analytical equipment supports complete blood cell counts (CBC) and analysis of serum components revealing metabolic or organ stress from small samples of mouse blood.

The facility also include a Tumor Analysis service (link)which provides expertise in analysis animal models of human cancer and other diseases. Investigators have access to a wide variety of human cancer cell lines for xenograft studies, primary human xenograft models as well as transgenic mouse solid tumors and leukemia models.

Services

Imaging and analytical services can be done by trained animal facility specialists. Training on the various instruments can also be provided for regular users. We also provide Luciferin and luciferase-labeled cell lines.

Tumor Analysis services is a resource of materials, expert advice and practical training for analyzing animal models of human diseases.

Equipment and Resources

IMAGING LAB

  • VisualSonics Vevo 770 (VisualSonics): High resolution in vivo ultrasound imaging system includes: Pulsed Wave Doppler, M-Mode, Contrast Software Mode, and 3D Acquisition and Visualization Package. Accessories: Ultrasound Guided Injection System, Anesthesia System and Advanced Physiological Monitoring System. The ultrasound is useful for visualization of embryos in early stage pregnancy; visualization of embryos for organ specific injection in utero; volume measurement of mouse mammary tumors for drug studies with fewer animals; volume measurements of mouse prostate tumors to evaluate tumor development without killing of the animals; potential to image pancreatic tumors; and with the potential for measuring tumor blood flow with contrast agents.
  • Xenogen IVIS 200 Imaging System (Xenogen): For non-invasive in vivo imaging of luminescence or fluorescence. Anesthesia system allows analysis of live mice. Use luciferase labeled tumor cells or transgenic reporter mice.

    We have 9 luciferase labeled cell lines:
    • Breast: MDA-MB-231-D3H2LN (the H2 line generates tumors faster than the standard MB-231 cells
    • Prostate: PC-3M-luc-C6 (a metastatic PC-3 subline)
    • Lung: A549-Luc-C8
    • Colorectal: HT-29-luc-D6
    • Cervical: HeLa-luc
    • Ovarian: SKOV3-luc-D3
    • Melanoma: B16-F10-Luc-G5 (our only mouse cell line, derived from C57BL/6 mice)
    • BxPc-3-luc2 (pancreas adenocarcinoma)
    • LoVo-6-luc-1 (colon adenocarcinoma)

    Transgenic reporter mouse: beta-Actin-luc mouse line from Xenogen called FVB/N-Tg(ß-Actin-luc)-Xen. This line is cryopreserved. For animal availability please contact Ling Wang at lwang@sanfordburnham.org.
  • Faxitron MX-20 (Faxitron X-ray Corporation): X-ray instrument for performing bone densitometry and body composition measurements using a dual-function digital and film imaging system.

ANIMAL FACILITY ANALYSIS LAB

  • VetScan HMII Hematology System (Abaxis): Measure 18-parameter blood count (CBC). This includes white blood cells, red blood cells, and platelets from 100 µl of whole blood.
  • VetScan VS2 (Abaxis): blood chemistries, electrolytes, blood gas and immunoassay instrument.
  • VetTest Chemistry Analyzer (IDEXX Laboratories): A blood and urine testing instrument that analyzes up to 12 biochemical tests simultaneously, using a single sample of serum, plasma, or urine, in about 6 minutes.
  • CODA-6 (Kent Scientific Corporation): non-invasive tail cuff blood pressure measurements for simultaneous analysis of 6 mice.
