Pouring Gradient Gels:

1. Use multi-caster (large gel) for Amersham Pharmacia SE 600 system.
2. Three large gels uses about 100 ml of gel volume.
3. Set-up gels in caster.
4. Test to make sure all tubing etc works.
5. For 50ml solution add 165ml of 10% APS and 16.5ml of TEMED.
6. Set pump speed at 48 (about 10 mls per minute)

   Clamp tubing when air bubbles begin forming in tubing towards end. Add water to mixer to prevent polymerization of acrylamide in the mixer chambers.
