Resources for the Maintenance, Manipulation and Image Acquisition of Stem Cells

XVivo Hypoxia Culturing System is a self-contained incubation system that allows one to culture, manipulate and visualize their cultures all under a user specified hypoxic or normoxic condition. link

Two Leica MZ6 Dissecting Stereomicroscope (Bright field/Dark field): These microscopes are housed in separate EnvirCo laminar flow hoods for the routine manipulation of stem cell cultures. link One of the systems also has connected a computer and camera system for image acquistion and is available for teaching hESC manipulation techniques.

Two Leica M165 FC Fluorescent Dissecting Stereomicroscopes & Camera System: These microscopes will be available for use for the routine maintenance of stem cell lines that contain fluorescent reporters. One will be housed in a 4 foot EnvirCo laminar flow hood and one will be housed in the XVivo Hypoxia System (See above). link

Leica DMI 4000B Inverted Fluorescent Microscope & Camera System This microscope will be available for image acquisition and will have available both the Metavue and the standard Leica image acquisition software along with a Hamamatsu Orca EG Camera. link

Olympus IX71 Inverted Fluorescent Microscope:  This microscope will be available for image acquisition and will have available both the Metavue and the standard Olympus image acquisition software. link

Applied Precision Deltavision Deconvolution System & Incubation Chamber: This system will be available for image acquisition and has Cytoshop software installed.

MVE 1830 Eterne Cryostorage System: This -190 degree vapor phase cell storage system has the capacity to store ~80,000 2 ml vials. This unit will have a back-up N2 source in addition to a 24-hour remote alarm. link

Roche Lightcycler 480, RT-PCR: This newly released Lightcycler from Roche allows one to perform reactions in either a 96- or 384- well format. The main feature, in addition to the high quality nature of Roche products, is the ease to switch from the 96- to 384- well format without having to recalibrate the instrument. link

Invitrogen Countess-Automated Cell Counter: Cassettes are available for purchase in the SCC.

 Invitrogen Neon Electroporation System:  This sytsem utilizes a single resuspension buffer not dependant on the cell type. The voltage, pulse width and pulse number are changeable. Reagents and tips are available for purchase in the SCC.

Two 6 foot and four 4 foot Tissue Culture Hoods for the general maintenance and manipulation of cultures .

Three 4 foot EnvirCo Laminar Flow Dissection Hoods for the manual manipulation of cultures . link

Fourteen Sanyo CO2 Tissue Culture Incubators which have a UV decontamination system that sterilizes the air as it passes through the chamber. link

Two Olympus CKX-41 Tissue Culture Scopes for the general visualization and basic image acquisition of cultures.

Three Sanyo upright -80 degree freezers: link

Three upright 4 degree 20.6 cu ft. refrigerators.

Three upright 20.6 cu ft. manual defrost freezers.
